Comparative responses of Paliurus ramosissimus explants under different in vitro culture media

Explants pretreated with 70% ethanol and 1% NaOCl were additionally treated with 5% PPM for 4 h and subsequently cultured on MS, WPM, or DKW basal media supplemented with antioxidants, activated charcoal, and plant growth regulators (NAA and BAP). Upper panels show culture responses on media without activated charcoal, while lower panels show responses on charcoal-supplemented media. Bacterial contamination was effectively suppressed by PPM pretreatment. Browning was observed in some control media, whereas media supplemented with antioxidants and activated charcoal markedly reduced phenolic oxidation and maintained tissue viability. DKW medium with activated charcoal supported improved tissue stability and shoot initiation.
